## Challenge

The full scope of medication burden in MASLD patients, including how polypharmacy risk differs by liver disease stage, sex, and comorbidity profile, had not been systematically characterized in a real-world, longitudinal population. Cross-sectional or trial-based cohorts lacked the breadth of concomitant medication data needed to assess drug to drug interaction risk at the population level.

## Solution

Target RWE's persistent longitudinal infrastructure within TARGET-NASH enabled continuous capture of medication records, comorbidity profiles, and disease severity data across academic and community sites, allowing a multivariate analysis of polypharmacy patterns over time that no single-site or trial-based dataset could replicate.

## Impact

By quantifying how polypharmacy risk scales with MASLD severity and differs by sex, this study equips drug developers designing MASLD trials to anticipate concomitant medication interference, informs safety surveillance programs, and flags the subset of patients most at risk for compound adverse effects from new therapies.
